Sam Acheampong was born in Brampton, Ontario, on January 12, 1996. Initially, he grew up playing basketball at Cardinal Ambrozic Catholic Secondary School and was encouraged to play football by football coaches. As a 14-year-old who was 6' 3" tall and around 240 pounds, he had the potential to use his inborn physique to build a career as an athlete, and so he did.

During his high school years, the Brampton native played as an offensive lineman but wanted to switch to a defensive lineman. Due to this, Sam chose Wilfrid Laurier University, among many others, as he believed Laurier had a better history of producing great defensive linemen. So Acheampong began his journey as a collegiate football player at Wilfrid in 2015 and finished in 2019 with a record of 53 tackles, 11.5 sacks, and a forced fumble in 31 outings.

Later in 2021, Acheampong was selected 20th overall by the Toronto Argonauts in the second round of the 2020 CFL Draft. However, his professional career started a bit late as the 2020 CFL season was canceled. He played his first professional game on August 7, 2021.
